Amphipol mediated surface immobilization of FhuA: a platform for label-free detection of the bacteriophage protein pb5.

Biotinylated amphipol was used to entrap FhuA (an E. coli outer membrane protein) and immobilize the FhuA-amphipol complex on streptavidin surfaces. Using this assembly, we have successfully devised surface-based assays for studying the recognition of FhuA by pb5 (a bacteriophage T5 protein) and determination of the affinity constant.

Optimization of 3D Surfaces of Dextran with Different Molecule Weights for Real-Time Detection of Biomolecular Interactions by a QCM Biosensor

Quartz crystal microbalance (QCM) has been extensively applied in real-time and label-free biomolecular interaction studies. However, the sensitive detection by QCM technology remains challenging, mainly due to the limited surface immobilization capacity. Here, a three-dimensional (3D) carboxymethyl dextran coated gold sensor chip surface was successfully fabricated with dextran of different mo...

A Conductometric Urea Biosensor by Direct Immobilization of Urease on Pt Electrode

Urease was immobilized on platinum electrode both by chemical binding and electropolymerization. The conductometric urea biosensor thus prepared showed a detection limit of 4.9×10-5 M and linear dynamic range from 4.9×10-5 to 5.8×10-3 M for urea concentration when the enzyme is covalently immobilized on Pt electrodes. Conductometric transducers respo...

Phospholipid biotinylation of polydimethylsiloxane (PDMS) for protein immobilization.

Polydimethylsiloxane (PDMS) surfaces can be functionalized with biotin groups by adding biotinylated phospholipids to the PDMS prepolymer before curing. The addition of beta-D-dodecyl-N-maltoside (DDM) in the solution blocks non-specific protein binding on these functionalized PDMS surfaces. We characterize the surface by measuring fluorescently labeled streptavidin binding. Single molecule tra...

Site-specific protein and peptide immobilization on a biosensor surface by pulsed native chemical ligation.

The site-specific immobilization of proteins and peptides on a surface represents a significant challenge because many proteomics applications require strategies that preserve the innate functionality of these biomolecules at interfaces. Unfortunately, routine protocols for protein immobilization usually result in the heterogeneous presentation of the ligand at the solid–liquid interface. As a ...
